Prem Swaroop Yadav

Harvard Medical School
USA View Profile

Research Interest:
To understand the molecular and genetic regulation of skeleton development usingmouse genetics as well as in vitro approaches. Although, significant progress has been made in understanding the role of major signaling pathways such as BMP, TGF-β, Wnt and Hedgehog during skeleton development. However, the molecular interaction of pathways and how they converge during skeleton development is not very well understood. Deciphering theseknowledgeswill further enhance our understanding about bone development and the pathophysiology of skeleton disorders such as osteoporosis and osteoarthritis. I am also interested in identification and characterization of novel small molecules agonist as well as antagonist of BMP and Wnt signaling pathwaysusing computational as well as in vitro assays. Such a small molecule can be used as a potential therapeutic agent in skeleton related ailments.

Profile:
Prem Swaroop Yadav have done Bachelor of Science (B.Sc.) in 2005 and Master of Science (M.Sc.) in 2007 both in biology major from Veer Bahadur Singh Purvanchal University, Jaunpur, India. He also did a Post Graduate Diploma in Computer Applications (PGDCA) in 2008 from Udai Pratap College Varanasi, India. He joined Indian Institute of Technology Kanpur (IITK) for my doctoral research in year 2009. In his PhD he studied the developmental biology of Bone and Cartilage development using both mouse as well as chicken as a model. He also I developed some in vitro tools. One of which is a reporter cell line “BRITER” for screening the small molecules agonist of BMP Signaling which is being sold by a USA based company Kerafast (www.kerafast.com/product/1986/bmpresponsive-reporter-osteoblast-cell-line-briter). In his PhD he conducted a RNA-Seq analysis using in house developed cell line which can be differentiated in to bone and cartilage. Using this approach he identified a set of novel BMP signaling downstream target genes to decipher the Gene Regulatory Network (GRN) downstream of BMP signaling (a major regulator of skeleton development) during endochondral bone development. During my doctoral research he got opportunities to get trained from some of the best laboratories. He learned mouse trans-genesis with Prof. Shinichi Aizawa at the centre for Developmental Biology in Japan. He worked with Prof. Jørgen Kjems to learn the RNA-Seq analysis at the Interdisciplinary Nanoscience Center (iNANO) at Aarhus University in Denmark. He moved to Harvard School of Dental Medicine (HSDM), Harvard Medical School, Boston in 2016 for my post-doctoral research. Currently he is studying the role Wnt/PCP signaling pathways during development of skeleton tissues. His research interest includes understanding at the molecular mechanism using mouse genetics to uncover the genetic regulation of skeleton development. This knowledge will eventually enable us to do better management of pathophysiology of skeleton disorders such as delayed fracture repair, osteoporosis and osteoarthritis. He is also interested in identifying the small molecules agonist and antagonist of BMP and Wnt signaling pathways, using computational modelling as well as in vitro assays, which can be used to cure the skeleton disorders.
